The present invention relates to a method for identifying or determining a compound that inhibits or reduces internal ribosome entry site (IRES) mediated translation. For example, the present invention provides a method for determining a compound that inhibits IRES-mediated translation, said method comprising expressing in a cell a counter selectable marker operably under the control of an IRES. A candidate compound is then introduced into the cell or contacted with the cell and the cell maintained under conditions that select against a cell expressing the counter-selectable marker gene. Accordingly, a cell in which IRES-mdiated translation of the counter-selectable reporter gene is selected, thereby identifying a compound that inhibits IRES-mediated translation. The present invention also provides compounds identified by the method.
La présente invention concerne un procédé d'identification ou de détermination d'un composé qui inhibe ou réduit la traduction facilitée par le site de liaison ribosomique interne (IRES). Par exemple, la présente invention concerne un procédé de détermination d'un composé qui inhibe la traduction facilitée par l'IRES, ledit procédé comprenant l'expression dans une cellule d'un marqueur contre-sélectionnable fonctionnant sous la commande d'un IRES. Un composé candidat est alors introduit dans la cellule ou mis en contact avec la cellule et la cellule maintenue dans des conditions qui sélectionnent par rapport à une cellule exprimant le gène marqueur sélectionnable par un compteur. En conséquence, une cellule dans laquelle la traduction facilitée par l'IRES du gène reporteur sélectionnable par un compteur est sélectionnée, identifiant de ce fait un composé qui inhibe la traduction facilitée par l'IRES. La présente invention concerne aussi des composés identifiés au moyen de ce procédé.
