This application relates to the cloning of the cDNA encoding Osteoblast Specific Factor-2 (OSF2), and applications of it. OSF2 is shown to be encoded by the CBFA1 locus, but differs, in its 5'-terminus, from the original CBFA1 transcript. Osf2 mRNA is further shown to exist as two splice variants. The characterisation of the functional domains of OSF2 is also described, as well as the phenotype of OSF2 transgenic animals. OSF2 was previously identified in osteoblast cell nuclear extracts, as a specific activity binding to OSE2, one of two Osteoblast Specific promoter Elements identified in the osteocalcin gene promoter.
