Nucleic acid and protein sequences relating to a cation channel which is sperm- specific (CatSper1) are disclosed. The CatSper1 protein is shown to be specifically expressed in sperm and to be necessary for sperm motility. Nucleic acids, vectors, transformed cells, transgenic animals, polypeptides, and antibodies relating to the CatSper1 gene and protein are disclosed. Also provided are methods of in vitro fertilization and contraception, methods of identifying modulators of CatSper1 activity, methods of genotyping subjects with respect to CatSper1, and methods of diagnosing and treating CatSper1- mediated disorders, including infertility.

Cette invention se rapporte à des séquences d'acide nucléique et de protéine correspondant à un canal cationique qui est spécifique du sperme (CatSper1). Il s'est révélé que cette protéine CatSper1 s'exprime spécifiquement dans le sperme et qu'elle est nécessaire à la motilité du sperme. Des acides nucléiques, des vecteurs, des cellules transformées, des animaux tansgéniques, des polypeptides et des anticorps relatifs à ce gène et à cette protéine CatSper1 sont décrits. Cette invention concerne également des procédés de fertilisation et de contraception <i>in vitro</i>, des procédés pour identifier les modulateurs de l'activité de CatSper1, des procédés de génotypage de sujets par rapport à CatSper1, ainsi que des procédés de diagnostic et de traitement des troubles induits par CatSper1, tels que l'infertilité.
