The present invention provides methods of predicting protein aggregation and designing aggregation inhibitors. One such method for predicting potential protein aggregation inhibiting peptide sequences, includes the steps of: a) identifying a peptide sequence forming at least part of an aggregation region in a target protein; b) testing whether said peptide sequence forms part of a .beta.-sheet; c) if a positive result is achieved in step b), extracting the adjacent strands of that sheet; d) identifying residues in the adjacent strands to said peptide sequence whose side chains interact with said peptide sequence, those residues forming a potential protein aggregation inhibiting peptide sequence. The present invention also provides methods of designing compounds using the residues identified in the above method; compounds produced by the methods and computer programs for carrying out the above methods.
La présente invention concerne des procédés permettant de prédire l'agrégation d'une protéine et de concevoir des inhibiteurs d'agrégation. Ce procédé permettant de prédire des séquences peptidiques potentielles inhibitrices de l'agrégation d'une protéine, inclut les étapes consistant à : a) identifier une séquence peptidique formant au moins une partie d'une région d'agrégation dans une protéine cible; b) tester si ladite séquence peptidique forme une partie d'un feuillet .beta.; c) extraire les brins adjacents à ce feuillet si un résultat positif est obtenu dans l'étape b); d) identifier les résidus des brins adjacents à ladite séquence peptidique dont les chaînes latérales interagissent avec ladite séquence peptidique, ces résidus formant une séquence peptidique potentielle inhibitrice de l'agrégation d'une protéine. La présente invention concerne également des procédés de conception de composés utilisant les résidus identifiés dans le procédé précédent; les composés produits par les procédés et les programmes informatiques permettant de mettre en AEuvre les procédés précédents.
