A method for detecting and enumerating viable microorganisms in a sample suspected of containing said microorganisms comprising: (1) contacting said sample with a cell nutritive resource and a cellular proliferation inhibitor, (2) contacting said sample with at least one fluorescence labelled oligonucleotidic probe able to specifically hybridise at least one portion of ribosomal nucleic acids of said microorganisms, (3) detecting and quantifying the fluorescent signal, in which the microorganisms are of the species Legionella pneumophila and in which the cellular proliferation inhibitor is selected from the group consisting of ciprofloxacin and cephalexin. The invention also includes a kit comprising: (1) a cell nutritive resource, (2) a cellular proliferation inhibitor, (3) at least one fluorescence labelled oligonucleotidic probe able to specifically hybridise at least one portion of ribosomal nucleic acids of said microorganisms, (4) a means for detecting and quantifying the fluorescent signal, in which the cellular proliferation inhibitor is selected from the group consisting of ciprofloxacin and cephalexin.
L'invention porte sur un procédé de détection et de dénombrement de microorganismes viables dans un échantillon supposé contenir lesdits microorganismes. Ce procédé consiste à (1) mettre léchantillon en contact avec une source nutritive de cellules et un inhibiteur de prolifération cellulaire; (2) mettre léchantillon en contact avec au moins une sonde oligonucléotidique marquée par fluorescence, apte à s'hybrider spécifiquement au moins à une partie d'acides nucléiques ribosomiques des microorganismes; (3) détecter et quantifier le signal fluorescent, les microorganismes étant des Legionella pneumophila et l'inhibiteur de prolifération cellulaire étant choisi dans le groupe constitué par la ciprofloxacine et la céphalexine. L'invention porte également sur un coffret comprenant (1) une source nutritive de cellules; (2) un inhibiteur de prolifération cellulaire; (3) au moins une sonde oligonucléotidique marquée par fluorescence, apte à s'hybrider spécifiquement au moins à une partie d'acides nucléiques ribosomiques des microorganismes; (4) des moyens pour détecter et quantifier le signal fluorescent, l'inhibiteur de prolifération cellulaire étant choisi dans le groupe constitué par la cyprofloxacine et la céphalexine.
