The present invention provides a polynucleotide which identifies and encodes a novel cysteine protease (NCP) expressed in cells of both the adrenal gland and human umbilical vein endothelium. The present invention also provides for antisense molecules designed from the nucleotide sequence or its complement. The invention further provides genetically engineered expression vectors and host cells for the production of purified NCP, antibodies or other molecules specifically binding to NCP and pharmaceutical compositions based on molecules which bind NCP. The invention specifically provides for diagnostic assays based on altered ncp expression and which allow identification of such a condition. These assays utilize probes designed from NCP encoding nucleic acid sequences or antibodies specifically binding NCP.
La présente invention se rapporte à un polynucléotide qui identifie et code une nouvelle cystéine protéase (NCP) exprimée à la fois dans les cellules des glandes surrénales et de l'endothélium de la veine ombilicale d'origine humaine. La présente invention se rapporte également à des molécules anti-sens conçues à partir d'une séquence nucléotidique de NCP ou de son complément. L'invention se rapporte en outre à des vecteurs d'expression obtenus par génie génétique et à des cellules hôtes destinées à la production de NCP purifiée, à des anticorps et à d'autres molécules se liant de manière spécifique à la NCP, de même qu'à des compositions pharmaceutiques à base de molécules qui se lient à la NCP. L'invention concerne particulièrement des méthodes de dosage diagnostique fondées sur l'expression de ncp non modifiée qui permettent l'identification de troubles liés à la ncp. Ces méthodes de dosage utilisent des sondes conçues à partir de séquences d'acides nucléiques codant la NCP ou d'anticorps se liant de manière spécifique à la NCP.
