The application provides novel neuroactive fragments of human amyloid precursor protein (APP). The fragments include sequence from the A.beta. region of APP but also extend upstream. The fragments are soluble in aqueous solution most bands are about 10 kDa and above and do not correspond with the expected molecular weight of a dimer or trimer. The fragments and sub fragments thereof can be used as immunogens in methods of immunotherapy to treat Alzheimer's and other amyloidogenic diseases characterized by deposits of A.beta. in the brain. Antibodies to the part of the fragment upstream from APP or the interface between A.beta. and the upstream sequence can also be used in such therapeutic method. The fragments are also useful as markers, which can be detected for diagnosis or prognosis of disease or for screening agents.
L'invention concerne de nouveaux fragments neuroactifs de protéine précurseur d'amyloïde humaine (APP). Les fragments comprennent une séquence depuis la zone Ab de APP, mais s'étendent également en amont. Les fragments sont solubles dans une solution aqueuse. La plupart des bandes sont d'environ 10 kDa et plus et ne correspondent pas au poids moléculaire attendu d'un dimère ou d'un trimère. Les fragments et leurs sous-fragments peuvent être utilisés en tant qu'immunogènes dans des procédés d'immunothérapie pour traiter la maladie d'Alzheimer et d'autres maladies amyloïdogéniques caractérisées par un dépôt de Ab dans le cerveau. Des anticorps à la partie du fragment en amont de l'APP ou à l'interface entre Ab et la séquence en amont peuvent également être utilisés dans un tel procédé thérapeutique. Les fragments sont également utiles en tant que marqueurs pouvant être détectés pour diagnostic ou pronostic d'une maladie ou comme agents de criblage.
