Potent neutralizing monoclonal antibodies to the human .alpha. PDGF receptor (a PDGFR) and fragments thereof are described. These monoclonal antibodies specifically bind to an epitope on .alpha. PDGFR, inhibit PDGF binding with PDGF receptor, antagonize PDGF, and do not bind .beta. PDGF receptor. A hybridoma cell line producing such a monoclonal antibody, methods of in vivo imaging of a pathological condition and methods of inhibiting dhe growth of a neoplasia expressing .alpha. PDGFR, which use these monoclonal antibodies are also described. In vitro assays for detecting dhe presence of .alpha. PDGFR and for evaluating the binding affinity of a test compound are also described.
L'invention concerne des anticorps monoclonaux de neutralisation puissants dirigés contre le récepteur du facteur de croissance dérivé des plaquettes alpha ( alpha PDGFR) et des fragments de ceux-ci. Ces anticorps monoclonaux se lient spécifiquement à un épitope sur alpha PDGFR, ils inhibent la liaison de PDGF avec PDGFR, ils ont une action antagoniste contre PDGF et ils ne se lient pas au récepteur de PDGF beta . Une lignée cellulaire d'hybridomes produisant un tel anticorps monoclonal est décrit, ainsi que des procédés d'imagerie in vivo d'un état pathologique ainsi que des procédés d'inhibition de la croissance d'un alpha PDGFR exprimant une néoplasie, dans lesquels sont utilisés ces anticorps monoclonaux. L'invention porte également sur des méthodes de dosage in vitro pour la détection de la présence de alpha PDGFR et pour l'évaluation de l'affinité de liaison d'un composé d'essai.
