A method for embryogenesis and a method for plant regeneration are disclosed. Microspore-containing plant segment from donor plants are harvested and incubated under pre-treatment conditions to maintain microspores at a uninucleate cell cycle G1 phase. Pre-treatment conditions comprise cold water or an aqueous solution of about 0.2 - about 1.0 mol/liter sugar alcohol, for example mannitol. Microspores are isolated from the plant segment and embryogenesis of microspores is induced in induction medium, thereby producing embryos. Green plants may be regenerated from the embryos produced. Arabinogalactan protein, auxin and ovary co-culture may be added to the induction medium to enhance embryogenesis from microspores.

L'invention concerne un procédé d'embryogenèse et un procédé de régénération de plantes. Des segments de plantes contenant des microspores de plantes donatrices sont ramassés puis incubés dans des conditions de pré-traitement afin de maintenir les microspores en phase G1 de cycle cellulaire uninucléé. Les conditions de pré-traitement comprennent de l'eau froide ou une solution aqueuse d'environ 0,2 à environ 1,0 mol/litre d'alcool, par exemple du mannitol. Les microspores sont isolés du segment de la plante et l'embryogenèse des microspores est induite dans un milieu d'induction, produisant ainsi des embryons. Les plantes vertes peuvent être régénérées à partir des embryons produits. On peut ajouter au milieu d'induction une co-culture de protéines d'arabinogalactane, d'auxines et d'ovaires afin d'améliorer l'embryogenèse à partir des microspores.
