Novel proline derivatives of the following formula <see fig.I> wherein: A is -O-, -NH-, <see fig.II> or absent; B is <see fig.III>, <see fig.IV>, <see fig.V>, <see fig.VI>, -(CH2)k- or <see fig.VII> where k is an integer of 1 to 3, R1 is hydrogen atom or a lower alkyl having 1 to 5 carbon atoms; W is <see fig.VIII>, <see fig.IX> or CH3- where R2 is hydrogen atom, halogen atom or a lower alkoxy; X is -S-, -SO-, -SO2-, -O- or -NH-; R is <see fig.X> or a lower alkyl having 1 to 5 carbon atoms where l is an integer of 0 to 3, Y and Z are the same or different and each is hydrogen atom, halogen atom, a lower alkyl having 1 to 5 carbon atoms which may be substituted by fluorine atom, amino, nitro, hydroxyl or a lower alkoxy, and Y and Z may combinedly form a saturated or unsaturated 5- or 6- membered ring; and n is an integer of 1 to 6, which specifically inhibit prolyl endopeptidase activity and can be used for the prevention and/or treatment of dementia and amnesia as agents which act directly on the central symptoms of dementia.

VI>, -(CH2)k- ou <voir fig. VII>, k est un entier de 1 à 3, R1 est un atome d'hydrogène ou un alkyle inf. de 1 à 5 atomes de carbone, W est <voir fig. VIII>, <voir fig. IX> ou CH3-, R2 est un atome d'hydrogène, un atome d'halogène ou un alcoxy inf., X est -S-, -SO-, -SO2-, -O- ou -NH-, R est <voir fig. X> ou un alkyle inf. de 1 à 5 atomes de carbone, l est un entier de 0 à 3, Y et Z sont identiques ou différents, chacun étant un atome d'hydrogène, d'halogène, un alkyle inf. de 1 à 5 atomes de carbone, avec ou sans substitution par un atome de fluor, un amino, un nitro, un hydroxyle ou un alcoxy inf., et Y et Z peuvent former en combinaison un cycle saturé ou insaturé de 5 ou 6 chaînons, enfin n est un entier de 1 à 6; ces dérivés sont des inhibiteurs spécifiques de l'activité de la prolylendopeptidase, et ils peuvent être employés pour la prévention et (ou) le traitement de la démence et de l'amnésie, en agissant directement sur les symptômes centraux de la démence.
