The present invention is directed to artificial antigen presenting cells and methods of making artificial antigen presenting cells. Such artificial antigen presenting cells may be used in certain methods of isolating and expanding T cell populations as well as modulating T cell responses. Additionally, the present invention provides novel methods for the identification and isolation of antigen-specific T cells. The methods provide for the construction of liposomes containing MHC:peptide complexes, accessory molecules, co- stimulatory molecules, adhesion molecules, and other molecules irrelevant to T cell binding or modulation that are used in the binding of artificial antigen presenting cells to solid support systems that may be used in the retrieval and identification of antigen-specific T cells. Additionally, the present invention is directed to devices and methods for treating conditions which would benefit from modulation of T cell response, for example, autoimmune disorders, allergies, cancers, viral infections, and graft rejection.

L'invention porte sur des cellules présentant des antigènes artificiels et leurs procédés d'obtention, lesdites cellules pouvant servir dans certains procédés d'isolement ou d'expansion de populations de cellules T, et à moduler la réponse des cellules T. L'invention porte également sur de nouveaux procédés d'identification et d'isolement de cellules T spécifiques d'antigènes consistant à construire des CMH contenant des liposomes: complexes de peptides, de molécules accessoires, molécules co-stimulatrices, molécules d'adhésion, et autres molécules étrangères à la fixation ou la modulation des cellules T, et servant à fixer les cellules présentant des antigènes artificiels à des systèmes supports solides pouvant eux-mêmes servir à détecter et identifier des cellules T spécifiques d'antigènes. L'invention porte en outre sur des dispositifs et procédés de traitement d'états pouvant bénéficier de la modulation de la réponse des cellules T, tels que par exemple des troubles auto-immunitaires, des allergies, des cancers, des infections virales, ou des rejets de greffes.
