Provided is a substantially modified cloning vector comprising a system that can be used for direct selection of recombinants through activation of antibiotic resistant gene. A fragment of the lacl gene involving in this system has been modified to introduce a multiple restriction enzyme digestion site. According to this invention, at least one antibiotic resistant marker is linked to and is under the control of a promoter that is regulated by lacl gene product. This structure is defined as lac-ABR operon which consists of lac promoter, lacl-regulated operator and antibiotic resistant marker coding sequence. Also, the vector contains a promoter named .DELTA.TAC that is derived from modification of TAC promoter and linked to lacl gene. With this promoter, lacl gene can be expressed permanently with high level. Any DNA can be introduced to the multiple cloning site (MCS) of the lacl DNA sequence by ligation to any restriction enzyme site within the MCS sequence.
