Novel piperidinyl compounds of formula (I) in which X and Y are are independently selected from hydrogen, halogen and C1-6 alkoxy, R1 is selected from five- and six- membered Image (I) heteroaryl rings containing from 1-3 heteroatoms selected from nitrogen, oxygen, and sulfur, and benz derivatives thereof, which may bear 1-2 substituents selected from C1-6 alkyl, hydroxy, C1-6 alkoxy which may bear a trifluoromethyl group, C1-6 alkoxycarbonyl, C1-6 hydroxyalkyl, benzyloxy, halo, C1-3 alkylaminocarbonyl, C1-3 alkyl, aminocarbonyl having the formula: CONRcRd wherein Rc and Rd are independently selected from hydrogen, 2-pyrrolidinyl, and C1-6 alkyl or wherein Rc and Rd together with the nitrogen atom to which each is attached form a 5- or 6-membered heterocyclic ring in which said nitrogen is the only heteroatom, ReS(O)n, RfNH, and RgS wherein Re and Rf are independently selected from hydrogen and C1-6 alkyl and n is 0, 1 or 2 and Rg is selected from C1-3 alkylcarbonylaminophenyl and di(C1-3)alkylamino(C1- 6)alkyl wherein at least one of the substituents on the five- and/or six-membered heteroaryl ring is a chiral sulfoxide of the formula C1-4 alkylSO, and their pharmaceutically acceptable salts, are useful as dopamine antogonists and are particularly useful as antipsychotic agents in humans. The compounds may also be useful in other neurological and psychiatric disorders including schizophrenia. Processes for preparing the piperidinyl compounds are also described as well as their use in medecine.
Nouveaux composés de pipéridinyle représentés par la formule (I), dans laquelle X et Y sont sélectionnés indépendemment parmi hydrogène, halogène et alcoxy C1-6, R1 est sélectionné parmi des noyaux d'hétéroaryle à cinq et six constituants contenant de 1 à 3 hétéroatomes sélectionnés parmi azote, oxygène et soufre, ainsi que leurs dérivés benz pouvant porter 1-2 substituants sélectionnés parmi alkyle C1-6, hydroxy, alcoxy C1-6 pouvant porter un groupe trifluorométhyle, alcoxycarbonyle C1-6, hydroxyalkyle C1-6, benzyloxy, halo, alkylaminocarbonyle C1-3, alkyle C1-3, aminocarbonyle possédant la formule: CONR<c>R<d> où R<c> et R<d> sont sélectionnés indépendamment parmi hydrogène, 2-pirrolidinyle, et alkyle C1-6, où R<c> et R<d>c, avec l'atome d'azote auquel chacun est fixé, constituent un noyau hétérocyclique à 5 ou 6 constituants dans lequel ledit azote est le seul hétéroatome, R<e>S(O)n, R<f>NH et R<g>S, où Re et R<f> sont sélectionnés indépendamment parmi hydrogène et alkyle C<1-6> et n est 0, 1 ou 2 et R<g> est sélectionnée parmi alkylcarbomylaminophényle C1-3 et di(C1-3)alkylamino(C1-6)alkyle, où au moins un des substituants du noyau hétéroaryle à cinq et/ou six constituants est un sulfoxyde chiral représenté par la formule C1-4alkylSO, ainsi que leurs sels acceptables pharmaceutiquement, présentant une efficacité en tant qu'antagonistes de dopamine et, particulièrement, en tant qu'agents antipsychotiques chez l'homme. Ces composés peuvent également être efficaces dans d'autres maladies neurologiques et psychiatriques, y compris la schizophrénie. L'invention concerne également des procédés de préparation de ces composés de pipéridinyle, ainsi que leur utilisation en médecine.
