Novel cap analogs which are easily synthesized, resulting in high levels of capping efficiency and transcription and improved translation efficiencies are provided. Such caps are methylated at the N7 position of one or both guanosines of the dinucleotide cap as well as at the 3' position on the ribose ring. Substituent groups on the ribose ring also result in the cap being incorporated in the forward orientation. Also provided are methods useful for preparing capped analogs and using mRNA species containing such analogs are also contemplated herein, as well as kits containing the novel cap analogs.
L'invention porte sur de nouveaux analogues de coiffes faciles à synthétiser permettant des niveau élevés de rendement de coiffage et de transcription et de meilleurs rendements de traduction. Ces coiffes sont méthylées à la position N7 d'une ou des deux guanosines de la coiffe de dinucléotide ainsi qu'à la position 3' du cycle de la ribose. Du fait de la présence des groupes substituants sur le cycle de la ribose, la coiffe est incorporée dans le sens vers l'avant. L'invention porte également sur des procédés de préparation d'analogues coiffés et d'utilisation d'espèces d'ARNm contenant lesdits analogues, et sur des trousses contenant les nouveaux analogues de coiffes.
