A method and apparatus are provided for optimizing a liquid chromatography/mass spectrometry system for gradient elution. A temperature profile is determined according to which a gas is flowed into an atmospheric pressure ionization interface for vaporizing a matrix flowed into the interface from a liquid chromatography column. A flow profile is determined according to which the gas is flowed into the interface for preventing entry of the matrix into the mass spectrometer when the matrix does not include an analyte for which detection is desired. The determination of the temperature and flow profiles is based on the characteristics of the matrix as it eludes from column into the interface. A heat control device and a flow control device can be provided for implementing the temperature profile and flow profile, respectively. The heating control device and flow control device can be programmed or controlled by software.
L'invention concerne un procédé et un dispositif servant à optimiser un système de spectrométrie de masse et de chromatographie liquide pour une élution graduée. On détermine un profil de température selon lequel un gaz est introduit dans une interface d'ionisation sous pression atmosphérique servant à vaporiser une matrice introduite dans l'interface depuis une colonne de chromatographie liquide. On détermine un profil d'écoulement selon lequel le gaz est introduit dans l'interface afin d'empêcher l'entrée de la matrice dans le spectromètre de masse quand cette matrice ne contient aucune substance à analyser dont on souhaite effectuer la détection. La détermination de ces profils de température et d'écoulement est basée sur les caractéristiques de la matrice simultanément à son élution depuis la colonne jusqu'à l'intérieur de l'interface. Un dispositif de régulation de chaleur et un dispositif de régulation d'écoulement peuvent contribuer à mettre en oeuvre les profils respectifs de température et d'écoulement. Ce dispositif de régulation de chaleur et ce dispositif de régulation d'écoulement peuvent être programmés ou contrôlé par logiciel.
