The present invention provides a method of detecting active antibody production in a blood sample, said method comprising contacting, in the presence and absence of protein synthesis inhibitor, aliquots of said sample with a solid phase under conditions which permit antibody production and secretion by the lymphocytes; detecting in solution, binding of antibody to said antigen(s) on the solid phase; and comparing said antibody binding in the presence or absence of protein synthesis inhibitor, whereby to obtain a determination of the amount of active antibody secretion in response to said antigen(s); and kits for performing the method. The method may also be modified to allow detection of non-specific infection indicators.
La présente invention concerne un procédé permettant de détecter la production active d'anticorps dans un échantillon de sang, ce procédé consistant premièrement à mettre en contact, en présence et en l'absence d'un inhibiteur de synthèse de protéines, des aliquotes dudit échantillon avec une phase solide dans des conditions qui permettent la production et la sécrétion d'anticorps par les lymphocytes; deuxièmement, à détecter, dans la solution, la liaison de l'anticorps avec ledit ou lesdits antigène(s) dans la phase solide, et enfin à comparer la liaison de l'anticorps en présence ou en l'absence d'un inhibiteur de synthèse de protéines, de façon à obtenir une détermination de la quantité d'anticorps actifs sécrétés en réponse audit ou auxdits antigènes. Cette invention concerne également des trousses de mise en oeuvre du procédé selon l'invention. Enfin, il est possible de modifier ce procédé pour permettre la détection d'indicateurs d'infections non-spécifiques.
