To date phylogeny has been used to compare entire families of proteins based on their nucleotide or amino-acid sequence. The present invention is directed to a novel analytical platform allowing the systematic comparison of families of proteins based on their biochemical properties utilizing high throughput assays. Integration and hierarchical clustering of the results for the Rho subfamily of GTPases revealed unexpected features of the small Rho GTPases when compared to amino-acid sequence-based trees. A novel phylo-biochemical classification of the Ras superfamily of GTPases is proposed.
