A method of releasing an agent (e.g. a chemotherapeutic) under predetermined conditions comprising the steps of protecting the agent within a lipid structure (e.g. a liposome), causing lipase activity to be constituted by combining two or more components (e.g. recombinant N- or C-terminal Clostridium perfringens alpha-toxin fragments), one of these components being conjugated to a targeting molecule (e.g. an antibody) which binds to a target (e.g. a tumour antigen) under the the predetermined conditions. The lipid structure is then exposed to the constituted lipase activity such as to release the agent. Also disclosed are materials and kits for use in the method.

Une méthode de libération d'un agent (p. ex. un agent chimiothérapeutique) dans des conditions prédéterminées comprenant les étapes suivantes : protection de l'agent avec une structure lipidique (p. ex. un liposome), faisant en sorte que l'activité de la lipase soit provoquée par la combinaison de deux constituants ou plus (p. ex. fragments N- ou C-terminaux de la toxine alpha de Clostridium perfringens recombinant), un de ces constituants étant conjugué à une molécule de ciblage (p. ex. un anticorps) qui se fixe à une cible (p. ex. un antigène tumoral) dans des conditions prédéterminées. La structure lipidique est ensuite exposée à l'activité de la lipase provoquée de manière à libérer l'agent. On divulgue également des matières et des trousses pour l'utilisation de la méthode.
