2064068 9102740 PCTABS00003 The present invention relates to methods and compositions for isolation of nucleic acids from cells. In particular aspects, this invention relates to the use of chaotropic compositions, such as guanidine hydrochloride or guanidinium isothiocyanate, in combination with polyanionic compositions, such as those containing sulfated polysaccharides (i.e., heparin or heparitin sulfate), for the isolation of nucleic acids (RNA and DNA). This method involves disrupting and lysing cells using a nucleic acid releasing composition containing a chaotropic component for the release of nucleic acids from the cell (guanidine hydrochloride or guanidinium isothiocyanate). The released nucleic acids are collected by ethanol precipitation and resuspended before exposure to a polyanion-containing protein dissociating composition which promotes the dissociation of nucleic acid associated proteins from the suspended nucleic acids. The isolated nucleic acids are washed, further collected by ethanol precipitation and resuspended in a selective buffer prior to further use.
