The present invention provides a novel isolated plasmid, wherein the plasmid is a native plasmid found in unique C. botulinum type A strains and encode either BoNT/ A3 or BoNT/ A4 and BoNT/B. The present invention also provides a method of obtaining a plasmid-encoded botulinum neurotoxin and botulinum neurotoxin complex comprising the step of isolating a plasmid en-coding the cntA/A or cntA/B neurotoxin gene and genes encoding protein components of the toxin complex from a C. botulinum type A strain. The inventors performed comparative analyses of representative BoNT/A subtype strains by pulsed-field gel electrophore-sis (PFGE) and Southern hybridizations with probes specific for the BoNT/A and B genes, cntA/A and cntA/B. Unexpectedly, the inventors determined that the genes encoding BoNT/ A3 in the A3 strain, and BoNT/ A4 and BoNT/B in the A4 strain, are on plas-mids.
La présente invention porte sur un nouveau plasmide isolé, le plasmide étant un plasmide natif trouvé dans des souches uniques de C. botulinum type A et codant soit pour BoNT/ A3 soit pour BoNT/ A4 et BoNT/B. La présente invention porte également sur un procédé d'obtention d'une neurotoxine de botulinum codée par un plasmide et d'un complexe de neurotoxines de botulinum, comprenant l'étape consistant à isoler un plasmide codant pour le gène de neurotoxine cntA/A ou cntA/B et de gènes codant pour des composants de protéine du complexe de toxines provenant d'une souche de C. botulinum type A. Les inventeurs ont effectué des analyses comparatives de souches de BoNT/sous-type A représentative par électrophorèse sur gel à champ pulsé (PFGE) et hybridations de Southern avec des sondes spécifiques pour les gènes BoNT/A et B, cntA/A et cntA/B. De façon inattendue, les inventeurs ont déterminé que les gènes codant pour BoNT/ A3 dans la souche A3 et BoNT/ A4 et BoNT/B dans la souche A4 sont sur des plasmides.
