The present invention relates to the derivates of polypeptide Exendin-4 that can promote the secretion of insulin and are useful in the treatment of diabetes type 2. The amino acid sequence of the derivates is as below: His-Gly- Glu-Gly-Thr-Phe-Thr-Ser-Asp-Leu-Ser-Lys-Gln-X-Glu-Glu-Glu-Ala-Val-Y-Leu-Phe- Ile-Glu-Trp-Leu-Lys-Asn-Gly-Gly-Pro-Ser-Ser-Gly-Ala-Pro-Pro- Pro- Ser-Z. Wherein X is Arg, Leu or Ile, Y is His, Arg or Lys, Z is Arg-OH, -OH, -NH2 or Lys-OH. The derivates could be synthesized by chemical methods or produced by DNA recombinant methods.
