The present invention relates to methods useful for identifying compounds capable of specifically regulating actin polymerization, stress fiber formation or focal adhesion assembly by regulating G alpha12 and/or G alpha13 activity in cells involved in inflammatory responses, immune responses, allergic responses and neuronal responses. The invention also describes kits to perform these assays and methods to control disease related to such responses. The figure illustrates stress fiber formation in Swiss 3T3 cells microinjected with expression plasmids encoding constitutively activated Rho, G alpha12 and G alpha13 protein.

L'invention concerne des procédés utiles pour identifier des composés capables de réguler spécifiquement la polymérisation de l'actine, la formation de fibres de contrainte ou la formation d'un ensemble d'adhésion focale par la régulation de l'activité G alpha 12 et G alpha 13 dans les cellules qui sont le siège de réactions inflammatoires, immunitaires, allergiques et neuronales. L'invention concerne également des kits permettant de mettre en oeuvre ces tests et des procédés pour soigner des maladies se manifestant par ces réactions. La figure illustre la formation de faisceaux de fibres de contrainte dans les cellules suisses 3T3 transformées par des plasmides d'expression codant pour les protéines à activateur constitutif Rho, G alpha 12 et G alpha 12.
