As a molecular material for synthesizing a polynucleotide via reverse translation from a protein encoded by the polynucleotide, a reverse translation-mediating oligonucleotide characterized by having the following properties (a) to (d) is provided: (a) containing a nucleotide sequence nnn encoding amino acid residues Xaa; (b) the nucleotide sequence nnn being separable; (c) containing a nucleotide sequence binding to the amino acid residues Xaa; and (d) being free from any nucleotide sequence binding to amino acid residues other than Xaa. Also, a method of synthesizing a polynucleotide which encodes the amino acid sequence of a template protein via a reverse translation reaction with the use of the above oligonucleotide is provided.
L'invention concerne un matériau moléculaire destiné à la synthèse d'un polynucléotide par traduction inverse, à partir d'une protéine codée par le nucléotide, ce matériau, un oligonucléotide assurant la médiation de traduction inverse, étant caractérisé par le fait (a) qu'il contient une séquence nucléotidique nnn codant pour des résidus acides aminés Xaa, (b) que cette séquence nnn n'est pas séparable, c) qu'il contient une séquence nucléotidique qui se lie aux résidus acides aminés Xaa, et (d) qu'il ne contient aucune autre séquence nucléotidique qui se lierait à des résidus acides aminés autres que Xaa. L'invention concerne aussi un procédé de synthèse d'un polynucléotide qui code pour la séquence d'acides aminés d'une protéine matrice par une réaction de traduction inverse au moyen de l'oligonucléotide décrit.
