The invention relates to an interacting polypeptide consisting of or comprising a heptapeptide pattern of sequence X¿1?X¿2?X¿3?X¿4?X¿5?X¿6?X¿7? and a transduction domain, characterized in that it is a chimera polypeptide, the amino acid X¿7? is located between 5 and 35 amino acids of the C-terminal end of said polypeptide, and that the domain (b) is situated in C-terminal relative to pattern (a). The invention also relates to screening methods for identifying interacting polypeptides capable of modifying the phenotype of a cell and to uses of interacting polypeptides as mentioned in phenotypic screens or for therapeutic purposes. Lastly, the invention concerns interacting polypeptides capable of modifying the function of the HIV-1 Rev viral protein.
La présente invention concerne un polypeptide d'interaction consistant en ou comprenant un motif heptapeptidique de séquence X1X2X3X4X5X6X7 et un domaine de transduction, caractérisé en ce qu'il s'agit d'un polypeptide chimère, que l'acide aminé X7 se trouve entre 5 et 35 acides aminés de l'extrémité C- terminale dudit polypeptide, et que le domaine (b) est situé en C-terminal par rapport au motif (a). Elle concerne également des procédés de criblage pour l'identification de polypeptides d'interaction capables de modifier le phénotype d'une cellule et des utilisations des polypeptides d'interaction tels que mentionnés dans des cribles phénotypiques ou pour des fins thérapeutiques. Enfin, la présente invention est dirigée vers des polypeptides d'interaction particuliers capables de modifier la fonction de la protéine virale Rev de HIV-1.
