The invention is directed to a method for treating a cyclooxygenase-2 mediated disease or condition in a mammalian patient at risk of a thrombotic cardiovascular event, wherein the patient is on aspirin therapy to reduce the risk of the thrombotic cardiovascular event, comprising orally concomitantly or sequentially administering to the patient a cyclooxygenase-2 selective inhibitor in an amount effective to treat the cyclooxygenase-2 mediate disease or condition, and a nitric oxide donating compound in accordance with Formula (I) or a pharmaceutically acceptable salt thereof, wherein the nitric oxide donating compound is administered in an amount effective to reduce the gastrointestinal toxicity caused by the combination of the cyclooxygenase-2 selective inhibitor and aspirin. Pharmaceutical compositions are also encompassed.
L'invention concerne une méthode permettant de traiter une maladie ou un état induit par la cyclooxygénase-2 chez un patient mammifère qui présente un risque d'événement cardio-vasculaire thrombotique, ledit patient étant soumis à une thérapie par aspirine afin de limiter ce risque. Ladite méthode consiste à administrer oralement à un patient, de manière concomitante ou séquentielle, un inhibiteur sélectif de la cyclooxygénase-2 en quantité efficace afin de traiter la maladie ou l'état induit par la cyclooxygénase-2, et un composé donneur d'oxyde nitrique représenté par la formule ou un sel pharmaceutiquement acceptable de celui-ci, ledit composé donneur d'oxyde nitrique étant administré en quantité efficace afin de réduire la toxicité gastro-intestinale entraînée par la combinaison de l'inhibiteur sélectif de la cyclooxygénase-2 et de l'aspirine. L'invention concerne également des compositions pharmaceutiques.
