The invention relates to a genetically engineered variant of a parent starch debranching enzyme, i.e. a pullulanase or an isamylase, the enzyme variant having an improved thermostability at a pH in the range of 4-6 compared to the parent enzyme and/or an increased activity towards amylopectin and/or glycogen compared to the parent enzyme, to methods for producing such starch debranching enzyme variants with improved thermostability and/or altered substrate specificity, and to a method for converting starch to one or more sugars using at least one such enzyme variant.

L'invention concerne un variant, obtenu par génie génétique, d'une enzyme parente de déramification de l'amidon, c'est-à-dire une pullulanase ou une isamylase, ce variant possédant une thermostabilité améliorée à un pH se situant entre 4 et 6 par rapport à l'enzyme parente et/ou une activité accrue à l'égard de l'amylopectine et/ou du glycogène par rapport à l'enzyme parente. L'invention concerne également des procédés de production de tels variants de l'enzyme de déramification de l'amidon, possédant une thermostabilité améliorée et/ou une spécificité de substrat modifiée, ainsi qu'un procédé de conversion de l'amidon en un ou plusieurs sucres, à l'aide d'au moins un tel variant d'enzyme.
