The present invention provides a process for making a compound having a structure according to formula (I) wherein A1, A2 and A3 are independently carbon or nitrogen and R1, R3, R4 and R6 are known quinolone substituents; and wherein one of R1, R3 or R6 may be a lactam-containing moiety; or a protected form, salt, pharmaceutically-acceptable salt, biohydrolyzable ester, or solvate thereof; the process comprising reacting one or more organosilicon compounds with a compound having a structure according to formula (II), wherein A1, A2 and A3, R1, R3, R4 and R6 as described above; wherein one of R1, R3 or R6 may be a lactam-containing moiety; and X is a leaving group; or a protected form, salt, biohydrolyzable ester, or solvate thereof. The compounds prepared according to the processes of the invention are themselves useful as antimicrobials, or they may be used as intermediates for making other quinolone-containing antimicrobials.
L'invention porte sur un procédé d'élaboration de composés antimicrobiens de structure conforme à la formule (I), dans laquelle A?1¿, A?2¿, et A?3¿ sont indépendamment carbone ou azote; et R?1¿, R?3¿, R?4¿ et R?6¿ sont des substituants connus de la quinolone, où l'un des éléments R?1¿, R?3¿, ou R?6¿ peut être une portion contenant du lactame. L'invention porte en outre sur une forme protégée, un sel, un sel pharmacocompatible, un ester biohydrolysable ou un solvate du composé de formule (I). Le susdit procédé consiste à faire réagir un ou plusieurs composés organosilicié avec un composé de structure conforme à la formule (II), dans laquelle A?1¿, A?2¿, et A?3¿ et, R?1¿, R?3¿, R?4¿ et R?6¿ sont comme ci-dessus, où l'un des éléments R?1¿, R?3¿ ou R?6¿ peut être une portion contenant du lactame, et où X est un groupe partant, ou avec une forme protégée, un sel, un sel pharmacocompatible, un ester biohydrolysable ou un solvate du composé de formule (II). Les composés préparés conformément au procédé de l'invention sont utiles comme antimicrobiens et peuvent servir d'intermédiaires pour la préparation d'autres antimicrobiens contenant de la quinolone.
