The invention provides methods and compositions for generating non-human transgenic animals that are humanized at one or more gene sequences. According to the methods of the invention, a DNA construct containing a human DNA sequence flanked by sequences from the non-human animal is generated by recombination in a bacterial cell, for example, in E. coli. The DNA construct that is produced can then be introduced into a non-human embryogenic stem cell where it can recombine with the genomic DNA of the non-human animal.

L'invention concerne des procédés et des compositions destinés à la production d'animaux transgéniques non humains, humanisés sur une ou sur plusieurs séquences de gènes. Selon les procédés de l'invention, une construction d'ADN contenant une séquence d'ADN humain flanquée de séquences de l'animal non humain est produite par recombinaison dans une cellule bactérienne, par exemple, dans E. coli. La construction d'ADN produite peut alors être introduite dans une cellule souche embryonnaire non humaine où elle peut se recombiner à l'ADN génomique de l'animal non humain.
