This invention relates to compounds of formula (I) wherein: X, Y and Z independently are N, O, S or CR', provided that at least two of X, Y and Z are heteroatoms and at least one of X, Y and Z is N; or one of X, Y and Z is C=N, C=C or N=N and the other two are CR' or N, provided that X, Y and Z together comprise at least two N; ~ indicates a single or double bond; R1 is R", R"C(O), R"C(S), R"SO2, R"OC(O), or R"OC(O)NR'CH(R6)C(O); R2 is H, C1-6alkyl, C2-6alkenyl, Ar-C0-6alkyl, or Het-C0- 6alkyl; R3 is H, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, C3-6cycloalkyl-C0-6alkyl, Ar-C0-6alkyl, or Het-C0-6alkyl; R4 is H C1-6alkyl, C2-6alkenyl, Ar-C0-6alkyl, or Het-C0-6alkyl; R5 is C1-6alkyl, Ar-C0-6alkyl, Het-C0-6alkyl, CH(R6)NR'R7, CH(R6)Ar, CH(R6)OAr, or NR8R9; R6, R7, R8 and R9 are as defined in the description; R10 is H, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, C3-6cycloalkyl-C0-6alkyl, Ar-C0- 6alkyl, or Het-C0-6alkyl; n is 1, 2 or 3; or a pharmaceutically acceptable salt thereof, which are inhibitors of cysteine porteases, particularly cathepsin K, and are useful in the treatment of diseases in which inhibition of bone loss is a factor.
