The invention relates to a method for the preparation of citalopram comprising, in either order, subjecting a compound of formula (III) wherein Y is a cyano group or a group which may be converted to a cyano group, R is hydrogen, -O-R1, NH2, NHCH3 or -N(CH3)2 wherein R1 is selected from hydrogen, alkyl, alkenyl, alkynyl and optionally alkyl substituted aryl or aralkyl; to i) reduction of the double bond in the side chain of formula -CH=CH-COR: ii) conversion of the group -COR or a reduced form thereof to a dimethylaminomethyl group; and iii) if Y is not cyano, conversion of the group Y to a cyano group; followed by isolation of citalopram base or a pharmaceutically acceptable acid addition salt thereof.
L'invention concerne un procédé servant à préparer citalopram et consistant, dans l'un ou l'autre ordre, à soumettre un composé représenté par la formule (III) dans laquelle Y représente un groupe cyano ou un groupe pouvant être converti en groupe cyano, R représente hydrogène, -O-R?1¿, NH¿2?, NHCH¿3? ou N(CH¿3?)¿2? dans laquelle R?1¿ est sélectionné dans hydrogène, alkyle, alkényle, alkynyle et, éventuellement, aryle ou aralkyle substitué par alkyle ; à (i) une réduction de la double liaison de la chaîne latérale représentée par la formule -CH=CH-COR ; (ii) une conversion du groupe -COR ou d'une de ses formes réduites en un groupe diméthylaminométhyle ; et (iii) si Y ne représente pas cyano, une conversion du groupe Y en un groupe cyano. On isole ensuite la base de citalopram ou un de ses sels d'apport acide acceptable sur le plan pharmaceutique.
