Abstract of the Disclosure The present invention relates to an improved process for synthesizing a peptide chain by reacting, in aqueous solution, an amino acid precursor complex containing a blocked alpha amino or carboxyl group and a polynucleotide handle with an amino acid segment, separating the reacted complex from unreacted acid by reversibly coupling the polynucleotide of the complex to a complementary polynucleo- tide adsorbent immobilized on an insoluble support and eluting the complex from the support as an aqueous solution. Preferably, the complex is enzymatically deblocked and used as the precursor for repeating the reaction with a further acid. Preferably, during the process, those chains which failed to react with a given acid are removed by enzymatic degradation.
