A high throughput screening method for identifying a compound that binds to a target species. The method includes: incubating a plurality of test mixtures under isothermal denaturing conditions, each test mixture including at least one test compound and at least one target species, wherein the isothermal denaturing conditions are effective to cause at least a portion of the target species to denature to a measurable extent. The method further involves detecting a denaturation signal of each target species in the presence of the at least one test compound; and comparing the denaturation signal of each target species in the presence of the at least one test compound with a denaturation signal of the same target species in the absence of the at least one test compound under the same isothermal denaturing conditions.

L'invention concerne un procédé de criblage à haut débit destiné à identifier un composé qui se lie à une espèce cible. Le procédé consiste notamment à incuber plusieurs mélanges test dans des conditions de dénaturation isothermes, chaque mélange comportant au moins un composé test et au moins une espèce cible. Les conditions de dénaturation isothermes sont efficaces pour provoquer la dénaturation dans des proportions mesurables d'au moins une partie de l'espèce cible. Le procédé consiste en outre à détecter un signal de dénaturation de chaque espèce cible en présence du composé test; et à comparer le signal de dénaturation de chaque espèce cible en présence d'au moins un composé test à celui de la même espèce cible en l'absence du composé pour essai dans les mêmes conditions de dénaturation isothermes.
