The present invention relates to compounds of the general formula wherein R1 is -(CH2)n-aryl or -(CH2)n-heteroaryl, wherein such groups are unsubstituted or substituted by one or more substituents, selected from the group consisting of halogen, lower alkyl substituted by halogen, lower alkoxy, lower alkyl, cyano, nitro, -O-lower alkyl substituted by halogen or morpholinyl; R2 is -(CH2)n-aryl or -(CH2)n-heteroaryl, wherein such groups are unsubstituted or substituted by one or more substituents, selected from the group consisting of lower alkyl, lower alkoxy, halogen or -N(lower alkyl)2; R3 is hydrogen or lower alkyl; R4 is -(CH2)n-aryl or -(CH2)n-heteroaryl, wherein such groups are unsubstituted or substituted by one or more substituents, selected from the group consisting of halogen or lower alkoxy, or is lower alkyl, -(CH2)n-cycloalkyl; or R3 and R4 form together with the N-atom a heterocyclic ring; n is 0, 1 or 2; and to pharmaceutically acceptable acid addition salts thereof. It has been found that the compounds of general formula I or their tautomeric forms are good inhibitors of the glycine transporter 1 (GIyT-1), and that they have a good selectivity to glycine transporter 2 (GIyT-2) inhibitors, useful for the treatment of schizophrenia.

La présente invention concerne des composés de formule générale où R1 représente un groupement -(CH2)n-aryle ou -(CH2)n-hétéroaryle, éventuellement substitué par un ou plusieurs substituants sélectionnés au sein du groupe constitué par les atomes d'halogène et les groupements alkyle inférieur substitués par un halogène, alcoxy inférieur, alkyle inférieur, cyano, nitro, -O-alkyle inférieur substitués par un halogène ou morpholinyle ; R2 représente un groupement -(CH2)n-aryle ou -(CH2)n-hétéroaryle, éventuellement substitué par un ou plusieurs substituants sélectionnés au sein du groupe constitué par les groupements alkyle inférieur, alcoxy inférieur, halogéno ou -N(alkyle inférieur)2; R3 représente un atome d'hydrogène ou un groupement alkyle inférieur ; R4 représente un groupement -(CH2)n-aryle ou -(CH2)n-hétéroaryle, éventuellement substitué par un ou plusieurs substituants sélectionnés au sein du groupe constitué par les atomes d'halogène et les groupements alcoxy inférieur, ou représente un groupement alkyle inférieur ou -(CH2)n-cycloalkyle ; ou R3 et R4 forment ensemble et avec l'atome d'azote un hétérocycle ; n est égal à 0,1 ou 2 ; ainsi que les sels d'addition acide de qualité pharmaceutique desdits composés. Il a été découvert que les composés de formule générale I ainsi que leurs formes tautomères sont de bons inhibiteurs du transporteur de glycine I (GlyT1), et qu'ils présentent une bonne sélectivité vis-à-vis des inhibiteurs du transporteur de glycine 2 (GlyT-2), employés dans le traitement de la schizophrénie.
