A process for obtaining 2'-halo-4-methylbiphenyls is described, which comprises reacting 4 halotoluene with a 1,2--dihalobenzene in the presence of elemental metal such as magnesium, lithium or zinc, wherein 0 to 0.9 molar, particularly 0 to 0.2 molar excess of 4-halotoluene in regard to 1,2-dihalobenzene is used, and arised organometal intermediates are quenched by ele-mental halogen. In addition, the coupling of arised 2'-halo-4- methylbiphenyls with 2-(1-propyl)-4-methyl-6-(1'-methylbenzimidazole-2- il)benzimidazole to afford 3'-(2'-halo-biphenyl-4-ylmethyl)-1,7'-dimethyl-2'-propyl-1H,3'H- [2,5']bibenzoimidazolyl, which can be further converted to organometallic compound and said organometallic compound is further reacted with formic acid derivative, such as N,N-dimethylformamide, alkylformiate or carbon dioxide to obtain telmisartan, is also described. Further de-scribed is use of in line analytics for monitoring the aforementioned reactions, process for preparing a pharmaceutical composition and/or dosage for, or use in preparing a medicament.
Linvention concerne un procédé pour obtenir des 2-halo-4-méthylbiphényles, lequel procédé comprend la réaction de 4-halotoluène avec un 1,2-dihalobenzène en présence de métal élémentaire tel que le magnésium, le lithium ou le zinc, un excès de 0 à 0,9 molaire, en particulier de 0 à 0,2 molaire, de 4-halotoluène par rapport au 1,2-dihalobenzène étant utilisé, et le refroidissement brusque des intermédiaires organométalliques produits par de l'halogène élémentaire. De plus, linvention décrit le couplage des 2-halo-4-méthylbiphényles produits avec du 2-(1-propyl)-4-méthyl-6-(1'-méthylbenzimidazole-2-yl)benzimidazole pour produire du 3'-(2'-halo-biphényl-4-ylméthyl)-1,7'-diméthyl-2'-propyl-1H,3'H-[2,5']bibenzoimidazolyle, que lon peut encore convertir en composé organométallique. Ledit composé organométallique est alors mis en réaction avec un dérivé de lacide formique, tel que le N,N-diméthylformamide, un formiate dalkyle ou du dioxyde de carbone pour obtenir du telmisartan. L'invention concerne en outre lutilisation de procédés danalyse en ligne pour surveiller les réactions susmentionnées, un procédé pour préparer une composition pharmaceutique et/ou un dosage pour un médicament ou lutilisation dans la préparation dun médicament.
