A method for the preparation of a bioadhesive compact matrix, which comprises the preparation of a uniform mixture of powders comprising one alkylcellulose or one hydroxy alkylcellulose and a non- water- soluble, water- swellable cross-linked polycarboxylic polymer; the preparation of compressed units starting from such powder mixture by direct compression and finally the heating of the compressed units thus obtained to a temperature in the range of 80 - 250°C for a time of 1-60 minutes; the powder mixture can also comprise at least one active substance and the compressed units thus obtained are characterized by a prolonged release, and have a release kinetics of the active substance substantially of zero order in an aqueous solution at pH 4-8.
L'invention porte sur un procédé pour la fabrication d'une matrice compacte bioadhésive, lequel comprend la préparation d'un mélange uniforme de poudres comportant une alkyl-cellulose ou une hydroxyalkyl-cellulose et un polymère poly-carboxylique réticulé pouvant gonfler à l'eau, non hydrosoluble; la préparation d'unités comprimées à partir d'un tel mélange de poudres par compression directe, et enfin le chauffage des unités comprimées ainsi obtenues à une température dans la plage allant de 80 à 250°C pendant une durée de 1 à 60 minutes. Le mélange de poudres peut également comporter au moins une substance active et les unités comprimées ainsi obtenues sont caractérisées par une libération prolongée et présentent une cinétique de libération de la substance active sensiblement d'ordre zéro dans une solution aqueuse à pH 4 à 8.
