Novel methods for the preparation of sertraline hydrochloride Forms II, III, V, VI, VII, VIII, IX and X. Novel Form II is also disclosed. According to the present invention, sertraline hydrochloride Form II may be produced directly form sertraline base or sertraline mandelate. It may also be produced from sertraline hydrochloride solvate and hydrate forms, and crystallized form new solvent systems. According to the present invention, sertraline hydrochloride form III may be produced by heating sertraline hydrochloride Forms V and VI. Sertraline hydrochloride Forms V and VI may be produced from either sertraline hydrochloride or sertraline base by crystallization. Sertraline hydrochloride Form VII may be produced by suspending sertraline chloride polymorph V in water, followed by filtration. Sertraline hydrochloride Forms VIII and IX may be produced by suspending sertraline base in water followed by acidification and filtration. Sertraline hydrochloride Form X may be produced by suspending sertraline hydrochloride in benzyl alcohol with heating, followed by filtration. Pharmaceutical compositions containing sertraline hydrochloride Forms II, III and V through X, and methods of treatment using such pharmaceutical compositions are also disclosed.
L'invention concerne de nouvelles méthodes de préparation de formes II, III, V, VI, VII, VIII, IX et X d'hydrochlorure de sertraline. Une nouvelle forme II est également décrite. Selon l'invention, la forme II d'hydrochlorure de sertraline peut être produite directement à partir d'une base de sertraline ou d'un mandélate de sertraline. Elle peut également être produite à partir de formes de solvate et d'hydrate d'hydrochlorure de sertraline, et cristallisée à partir de nouveaux systèmes de solvants. Selon l'invention, la forme III d'hydrochlorure de sertraline peut être produite par chauffage des formes V et VI d'hydrochlorure de sertraline. Les formes V et VI d'hydrochlorure de sertraline peuvent être produites soit à partir d'hydrochlorure de sertraline ou d'une base de sertraline par cristallisation. La forme VII d'hydrochlorure de sertraline peut être produite par le biais d'une suspension d'un polymorphe V de chlorure de sertraline dans de l'eau, suivie d'une filtration. Les formes VIII et IX d'hydrochlorure de sertraline peuvent être produites par le biais d'une suspension d'une base de sertraline dans de l'eau, suivie d'une acidification et d'une filtration. La forme X d'hydrochlorure de sertraline peut être produite par le biais d'une suspension d'hydrochlorure de sertraline dans de l'alcool benzylique associée à un chauffage et suivie d'une filtration. Des compositions pharmaceutiques contenant les formes II, III et V jusqu'à X d'hydrochlorure de sertraline, et des méthodes de traitement faisant appel à de telles compositions pharmaceutiques sont également décrites.
