An improved method for ex-vivo generation of tumor-reactive T cells is provided, comprising, for example, culturing PBMC from a patient with cancer with autologous tumor cells and magnetic beads that can activate T lymphocytes via CD3 in combination with CD28, CD40, or CD28 plus CD40. The invention also provides genes encoding anti-CD3 or anti-4~1BB scFv and methods to transfect cells of neoplastic or normal origin for expression of those genes at their surface. The genes and transfected tumor cells are useful compositions for induction of a tumor-destructive immune response.
La présente invention concerne une méthode améliorée de génération ex-vivo de cellules T réagissant aux tumeurs qui comprend par exemple, la culture de PBMC provenant d'un patient atteint d'un cancer, avec des cellules tumorales autologues et des billes magnétiques qui peuvent activer les lymphocytes T via CD3 en combinaison avec CD28, CD40, ou CD28 plus CD40. Cette invention concerne également des gènes codant scFv anti-CD3 ou anti-4-1BB et des méthodes de transfection de cellules d'origine néoplasique ou normale pour exprimer ces gènes au niveau de leur surface. Les gènes et les cellules tumorales transfectées constituent des compositions utiles pour induire une réponse immunitaire détruisant les tumeurs.
