The invention relates to lipopeptide building blocks consisting of a peptide chain comprising a coiled-coil domain, linked covalently to a lipid moiety comprising long alkyl or alkenyl chains, and optionally linked to an antigen; and to helical lipopeptide bundles and synthetic virus-like particles formed by aggregation. The nanometer size and shape of these bundles and particles, their stability under aqueous physiological conditions, their chemical composition, the possibility to incorporate B- and T-cell epitopes, and their production by chemical synthesis, make them highly suitable as vaccine delivery vehicles.
L'invention porte sur des blocs de construction de lipopeptides constitués d'une chaîne de peptide comprenant un domaine bispiralé, liés de façon covalente à une fraction lipidique comprenant de longues chaînes alkyle ou alcényle, et facultativement liés à un antigène ; et sur des faisceaux de lipopeptides hélicoïdaux et des particules de type virus synthétiques formées par agrégation. La dimension nanométrique et la forme de ces faisceaux et particules, leur stabilité dans des conditions physiologiques aqueuses, leur composition chimique, la possibilité d'incorporer des épitopes de lymphocytes B et T et leur production par synthèse chimique, les rendent hautement appropriés en tant que véhicules d'administration de vaccins.
