The invention relates to site directed drug delivery systems providing a controlled release of L-dopa, carbidopa, L-.alpha.-methyldopa and/or their combinations which eliminates or at least reduces the deleterious side effects of these drugs. Biologically active copolymers having directional characteristics to the digestive mucous tissue comprise L-dopa, carbidopa or L- .alpha.-methyldopa chemically combined with biologically acceptable poly(acrylic acid) or poly(methacrylic acid). The copolymers are obtained by a process comprising the steps of: (a) reacting L-dopa, carbidopa or L-.alpha.- methyldopa with a chlorination agent; (b) bringing into reaction the product of step (a) and poly(acrylic acid) or poly(methacrylic acid) in the presence of a suitable catalyst; (c) washing the copolymer obtained in step (b) with a suitable organic solvent and drying it until constant weight. Controlled release antihypertensive pharmaceuticals comprise at least one biologically active copolymer selected from the group consisting of poly(acrylic acid/L- .alpha.-methyldopa), poly(methacrylic acid/L-$G(a)-methyldopa), poly(acrylic acid/carbidopa), poly(methacrylic acid/carbidopa) or combinations thereof and a pharmaceutically acceptable vehicle. Controlled release pharmaceuticals comprising at least one biologically active copolymer selected from the group consisting of poly(acrylic acid/L-dopa), poly(methacrylic acid/L-dopa), poly(acrylic acid/carbidopa), poly(methacrylic acid/carbidopa) or combinations thereof and a pharmaceutically acceptable vehicle are used in the therapeutic treatment of Parkinson syndrome.
La présente invention concerne des systèmes d'administration de médicaments dirigés permettant de libérer de manière régulée L-dopa, carbidopa, L-.alpha.-méthyldopa et/ou leurs associations et d'éliminer, ou au moins réduire, les effets secondaires délétères de ces médicaments. Des copolymères actifs sur le plan biologique présentant des caractéristiques directionnelles par rapport à la muqueuse digestive comprennent L-dopa, carbidopa ou L-.alpha.-méthyldopa combinés chimiquement avec l'acide poly(acrylique) ou l'acide poly(méthacrylique) acceptables sur le plan biologique. On obtient ces copolymères par un processus consistant à: (a) faire réagir L-dopa, carbidopa ou L-.alpha.-méthyldopa avec un agent de chloration; (b) mettre à réagir le produit de (a) et l'acide poly(acrylique) ou l'acide poly(méthacrylique) en présence d'un catalyseur approprié; (c) laver le copolymère obtenu dans (b) avec un solvant organique approprié et le sécher jusqu'à obtenir un poids constant. Les produits pharmaceutiques hypotenseurs à libération contrôlée comprennent au moins un copolymère biologiquement actif choisi dans le groupe comportant l'acide poly(acrylique/L-.alpha.-méthyldopa), l'acide poly(méthacrylique/L-.alpha.-méthyldopa), l'acide poly(acrylique/carbidopa), l'acide poly(méthacrylique/carbidopa) ou des combinaisons de ceux-ci et un excipient acceptable sur le plan pharmaceutique. Ces produits pharmaceutiques à libération contrôlée comprenant au moins un copolymère actif sur le plan biologique choisi dans le groupe comprenant l'acide poly(acrylique/L-dopa), l'acide poly(méthacrylique/L-dopa), l'acide poly(acrylique/carbidopa), l'acide poly(méthacrylique/carbidopa) ou des associations de ceux-ci acceptables sur le plan pharmaceutique ainsi qu'un excipient acceptable sur le plan pharmaceutique, sont utilisés dans le traitement thérapeutique du syndrome de Parkinson.
