The present invention relates to phosphodiesterase (PDE) enzyme inhibitors and to their use in pharmaceutical articles of manufacture. In particular, the present invention relates to potent inhibitors of cyclic guanosine 3',5'- monophosphate specific phosphodiesterase type 5 (PDE5) that when incorporated into a pharmaceutical product at about 1 to about 10 mg unit dosage are useful for the treatment of sexual dysfunction by daily administration of the PDE5 inhibitor. The articles of manufacture described herein are characterized by PDE5 inhibition, and accordingly, provide a benefit in therapeutic areas where inhibition of PDE5 is desired, especially erectile dysfunction, with minimization or elimination of adverse side effects resulting from inhibition of other phosphodiesterase enzymes and with an improvement of vascular conditioning.

La présente invention porte sur des inhibiteurs de l'enzyme phosphodiestérase (PDE) et sur leur utilisation dans la fabrication de produits pharmaceutiques. Cette invention porte notamment sur de puissants inhibiteurs de la phosphodiestérase de type 5 (PDE5) spécifique de la guanosine 3',5'-monophosphate cyclique qui, lorsqu'ils sont introduits dans un produit pharmaceutique en dose unitaire d'environ 1 à environ 10 mg, sont utiles dans le traitement de la dysfonction sexuelle, l'inhibiteur PDE5 étant administré quotidiennement. Ces produits précités se caractérisent en ce qu'ils inhibent PDE5, et en conséquence, ont des résultats avantageux dans les zones thérapeutiques où est souhaitée l'inhibition de PDE5, et notamment de la dysérection, et minimisent ou éliminent les effets secondaires résultant de l'inhibition d'autres enzymes phosphodiestérases et améliorent l'état vasculaire.
