An enzyme capable of catalyzing the removal of nitrite from pentaerythritol tetranitrate (PETN) is provided. The enzyme (known as PETN reductase enzyme) is produced by culturing a novel strain of the Enterobacter cloacae bacterium isolated from nature. The strain designated PB2 has been deposited as NCIMB 40718. The amino acid sequence of the enzyme and the genetic sequence which encodes for this enzyme have also been determined. A PETN reductase enzyme encoded by the on gene is provided. A method for producing PETN reductase enzyme in large quantities and methods of bioremediation using the enzyme so produced are also provided. Additionally there is provided a method of detecting the presence of PETN in a sample together with a biosensor for use in such a method.
L'invention concerne une enzyme capable de catalyser la suppression du nitrite contenu dans du pentaérythritol tétranitrate (PETN). On obtient cette enzyme (appelée PETN réductase) au moyen de la culture d'une nouvelle souche d'Enterobacter cloacae bacterium isolée du milieu naturel. On a déposé cette souche désignée PB2 en tant que NCIMB 40718. On a également déterminé la séquence d'acides aminés de l'enzyme, ainsi que la séquence génétique codant pour cette enzyme. L'invention concerne une enzyme PETN réductase codée par le gène onr. Elle concerne également un procédé de production de l'enzyme PETN réductase en quantités importantes, ainsi que des procédés de biodégradation accélérée au moyen de l'enzyme obtenue. Elle concerne, de plus, un procédé servant à détecter la présence de PETN dans un spécimen, ainsi qu'un biodétecteur servant à mettre ce procédé en application.
