This invention relates to methods of sequencing DNA. More specifically, this invention relates to methods of sequencing both the sense and antisense strands of DNA through the use of blocked and unblocked sequencing primers. In brief, these methods include the steps of annealing an unblocked primer to a first strand of nucleic acid; annealing a second blocked primer to a second strand of nucleic acid; elongating the nucleic acid along the first strand with a polymerase; terminating the first sequencing primer; deblocking the second primer; and elongating the nucleic acid along the second strand.

L'invention concerne des procédés de séquençage d'ADN. Plus précisément, elle concerne des procédés de séquençage des brins sens et des brins antisens d'ADN par utilisation d'amorces de séquençage bloquées et non bloquées. Ces procédés consistent notamment à anneler une amorce non bloquée avec un premier brin d'acide nucléique ; à anneler une seconde amorce bloquée avec un second brin d'acide nucléique ; à allonger l'acide nucléique le long du premier brin avec une polymérase ; à terminer la première amorce de séquençage ; à débloquer la seconde amorce ; et à allonger l'acide nucléique le long du second brin.
