A method for the measurement of an analyte in biological samples whereby an uncompetitive inhibitor is coupled to a ligand to form a ligand-inhibitor conjugate which is utilized in a homogeneous assay. The ligand-inhibitor conjugate has a structure: (see above formula) wherein: n = 1-2, and X is a drug derivative selected from the group of: phenytoin-3-butyramido-, gentamicin(C1a)-N1-carbonyloxy-, gentamicin(C1a)-N1-teraphthaloylamino-, 5'-[[[(digoxin(1,4-oxazepin))-4-yl]-phenyl-4-carbonyl]amino]-, and 5'-[[[(digitoxin(1,4-oxazepin))-4-yl]-phenyl-4-carbonyl]amino]-. The analyte can be a drug or drug derivative, hormone, polypeptide, or oligonucleotide. The present invention also provides the novel ligand-inhibitor conjugates, assay reagents and packaged kits useful for performing such measurements.
