Prion protein (PrP) peptides having at least one .alpha.-helical domain and forming a random coil conformation in aqueous solutions bind cellular PrP (PrPc) to form a complex having characteristics of the scrapie isoform (PrPSc). Methods for screening compounds able to inhibit or decrease the binding of PrP peptides to PrPc are disclosed, as well as methods for assaying PrPSc.
L'invention porte sur des peptides constitués de protéines prions (PrP) présentant au moins un domaine hélicoïdal .alpha., prenant dans une solution aqueuse une configuration aléatoire en hélice et qui peuvent se fixer aux Prp des cellules (PrP?c¿) pour former un complexe ayant les caractéristiques de l'isoforme de la tremblante du mouton (PrP?Sc¿). L'invention porte également sur des procédés de criblage de composés empêchant ou réduisant la fixation des peptides PrP sur les PrP?c¿ et sur des procédés d'analyse du PrP?Sc¿.
