To construct a system for producing an industrially useful substance through gene recombination by utilizing characteristics of a yeast, it is intended to provide a yeast in which a specific gene alone has been disrupted and to which auxotrophy is imparted. A yeast strain in which ADE1 gene has been disrupted is constructed by homologous recombination with a chromosomal DNA by using an ADE1 gene fragment encoding phosphoribosylaminoimidazole succinocarboxamide synthase (EC6.3.2.6) of <I>Candida maltosa</I> . Then, this disrupted strain is transformed by a gene expression cassette containing a biodegradable polyester synthase gene and the obtained transformant is cultured to thereby produce biodegradable polyester.
Pour élaborer un système de production industrielle d'une substance utile par recombinaison de gènes en utilisant les caractéristiques d'une levure, on cherché à obtenir une levure dont seul un gène spécifique à été interrompu et doté d'auxotrophie. La souche de levure dont le gène a été interrompu est élaborée par recombinaison homologue à l'aide d'un ADN de chromosome en utilisant un fragment du gène ADE1 codant pour la phosphoribosylaminomidazole succinocarboxamide synthase (EC6.3.2.6) du <i>Candida maltosa</i>; puis ladite souche interrompue est transformée par une cassette d'expression de gènes contenant un gène de synthase de polyester biodégradable; puis le transformant obtenu est mis en culture pour produire le polyester biodégradable.
