This invention concerns new methods and compositions that are useful in preventing and ameliorating cachexia. the clinical syndrome of poor nutritional status and bodily wasting associated with cancer and other chronic diseases. More particularly, the invention relates to aromatic guanylhydrazone (more properly termed amidinohydrazone) compositions and their use to inhibit the uptake of arginine by macrophages and/or its conversion to urea. These compositions and methods are also useful in preventing the generation of nitric oxide (NO) by cells, and so to prevent NO-mediated inflammation and other responses in persons in need of same. In another embodiment, the compounds can be used to inhibit arginine uptake in arginie-dependent tumors and infections.
Cette invention concerne de nouveaux procédés et de nouvelles compositions utiles dans la prévention et le traitement de la cachexie, qui est le syndrome clinique d'un mauvais état nutritionnel et de l'atrophie de parties du corps associé au cancer et à d'autres maladies chroniques. Plus particulièrement, l'invention concerne des compositions de guanylhydrazone aromatique (dont l'appellation correcte est amidinohydrazone) et leurs utilisations pour empêcher la fixation de l'arginine par des macrophages et/ou la transformation de celle-ci en urée. On peut également faire appel à ces compositions et à ces procédés pour prévenir la production d'oxyde nitrique (NO) par les cellules, et ainsi prévenir les inflammations induites par NO et d'autres réactions chez des personnes en manquant. Dans un autre mode de réalisation, les composés peuvent être utilisés pour inhiber la fixation de l'arginine dans des tumeurs et des infections dépendant de l'arginine.
