2087961 9202526 PCTABS00010 Comb-type branched polynucleotides of formula (I) are used as amplification multimers in nucleic acid hybridization assays, where S is a first spacer segment of at least about 15 nucleotides, X is a modified nucleotide that provides a branch site, S' is a branching site spacer segment of 0 to about 15 nucleotides, m is an integer equal to or greater than 15, R is a cleavable linker molecule, n is 0 or 1, S'' is a second spacer segment of about 0 to 10 nucleotides, A is a segment that is capable of hybridizing specifically to analyte nucleic acid or nucleic acid bound to analyte, S''' is a third spacer segment of 0 to 10, E is an oligonucleotide extension of 5 to 10 nucleotides, and L is a segment containing 2 to 10 iterations, of a sequence that is capable of hybridizing specifically to a labeled oligonucleotide probe.
