The invention relates to a method for in vitro determining thrombin activity in a sample wherein the sample is a blood sample and thrombin generation is measured by the steps of: - contacting a layer of said sample with a fluorogenic substrate of thrombin, wherein said layer has a thickness within a range of 0.05 to 5 mm and a surface within a range of 10 to 500 mm2; - allowing thrombin to generate in said sample; - measuring the fluorescence emitted from the surface of the layer, by the fluorescent group released from the fluorogenic substrate as a result of enzymatic action of generated thrombin on said fluorogenic substrate.
L'invention concerne un procédé permettant de mesurer in vitro l'activité de la thrombine dans un échantillon. L'échantillon est un échantillon de sang, et la production de thrombine est mesurée par les étapes consistant à appliquer une couche dudit échantillon sur un substrat fluorogène de la thrombine, ladite couche présentant une épaisseur comprise dans un intervalle de 0,05 à 5 mm, et une surface comprise dans un intervalle de 10 à 500 mm2, à permettre à la thrombine de se former dans ledit échantillon, à mesurer la fluorescence émise à la surface de la couche par le groupe fluorescent libéré par le substrat fluorogène sous l'effet de l'action enzymatique de la thrombine produite sur ledit substrat fluorogène.
