The present invention is related to a method for a simultaneous determination of the relative amounts of more than one target polynucleotide sequence and nucleotide variations in said targets. The method is carried out by separating and recording single-stranded probes, which have hybridized to the targets and which are determined and distinguished by their defined properties including size and optional detectable label. The probes are complementary to a region in the target that has a sequence being contiguous to the nucleotide variations to be determined. After being hybridized with affinity - tagged targets, the probes are attached to a solid support and purified. The target probe hybrids are elongated using enzyme - assisted elongations. The elongated pro bes are recorded after release from the solid supports and the amount of each of the targets and their nucleotide variations and the ratio of modified and modified target polynucleotide sequences are calculated from the recorded results. Also disclosed is a test kit, which kit comprises in a packaged form devices equipments and reagents as well as instructions for carrying out the method. The method is useful for several diagnostic purposes.
La présente invention concerne un procédé permettant de déterminer simultanément les quantités relatives de plus d'une séquence polynucléotidique cible et des variations nucléotidiques dans lesdites cibles. Le procédé consiste à séparer et à enregistrer des sondes simple brin, lesquelles se sont hybridées avec les cibles et qui sont déterminées et différenciées par leurs propriétés définies comprenant la taille et un marqueur détectable facultatif. Les sondes sont complémentaires à une région dans la cible qui a une séquence contiguë aux variations nucléotidiques à déterminer. Après hybridation avec les cibles marquées par affinité, les sondes sont fixées à un support solide et purifiées. Les hybrides cible/sonde sont allongés à l'aide d'allongements assistés par des enzymes. Les sondes allongées sont enregistrées après libération de leurs supports solides et la quantité de chacune des cibles et de leurs variations nucléotidiques ainsi que le rapport entre les séquences polynucléotidiques cibles modifiées et non modifiées sont déterminés à partir des résultats enregistrés. L'invention décrit également une trousse d'essai, ladite trousse comprenant sous forme de package des dispositifs, des appareils et des réactifs ainsi que les instructions permettant de mener à bien le procédé. Le procédé est utile à plusieurs fins de diagnostic.
