A method whereby a cDNA library having an extremely high full-length ratio can be constructed starting with a material in a very small amount compared with the conventional methods. This method comprises optimizing reaction conditions in the steps of the alkaline phosphatase reaction, acid pyrophosphatase reaction and RNA ligase reaction in the conventional oligocap method.
L'invention se rapporte à un procédé de création d'une banque d'ADNc comportant un pourcentage extrêmement élevé d'ADNc pleine longueur, et ce à partir d'une très faible quantité de matière en comparaison des procédés de construction classiques. Ledit procédé consiste à optimiser les conditions de réaction lors des étapes de réaction des phosphatases alcalines, de réaction des pyrophosphatases acides et de réaction des ARN ligases de la méthode oligocap traditionnelle.
